Plicamycin, also known as mithramycin or Mithracin, is an antineoplastic antibiotic produced by the bacterium Streptomyces plicatus. Plicamycin binds to DNA and inhibits both DNA and RNA synthesis. It accomplishes this by binding to the guanine-cytosine (GC) rich regions of DNA, preventing the transcription of genetic information into RNA. As a result, the synthesis of proteins and other essential cellular components is disrupted.
Plicamycin inhibits the activity of osteoclasts, the cells responsible for breaking down bone tissue. Also, it interferes with the function of certain transcription factors, such as Sp1 (specificity protein 1). Transcription factors are proteins that regulate the transcription of genes. By disrupting the activity of transcription factors, plicamycin affects the expression of genes involved in cell growth and differentiation.
Researchers may employ plicamycin to investigate its effects on gene expression, cell cycle regulation, and apoptosis in cancer cells.
